Kindly note that your plasmid should comply to current GMO and biosafety regulations and RIBOPRO will need to evaluate if your plasmid is suitable in our workflow.

7. In case we order the DNA template for you, would you like the polyA to be designed in the plasmid? And if yes, do you have any specific request for polyA length?

Kindly note that in case this is not needed, our standard product is with 150 nt polyA tail

8. Would you like us to de-immunize your sequence with our RIBOSTEALTH TM method?

RIBOSTEALTH TM involves the exchange of synonymous codons throughout the coding sequence to remove TLR7 and 8 motifs within the RNA. It enhances activity of the RNA by reducing its immunogenicity and creates a unique (patentable) sequence. Licensing fees will apply, please consult our Product Specialist for more information.

Our standard QC test for RUO RNA products includes: 1)assessing the purity of RNA using spectrophotometry (A260/280 > 2, A260/230 > 2.2) 2) evaluating the size and integrity of the RNA product through gel electrophoresis (with less than 5% by product). Additionally, upon request, we can perform a dsRNA Elisa to determine the percentage of dsRNA within the sample.

Our standard LNP formulation is developed by RIBOPRO (RUO grade) and consist of RIBOPRO’s: • Ionizable lipid L1 • DSPE-PEG2000 • DSPC • Our patented core lipid resveratrol (described in patent application GB2302736.0) • DOTAP Our standard formulation is ideal for therapeutic use because the ionizable lipid is low in toxicity and exhibits reduced off-target effects to the liver compared to cholesterol-based formulations.

4. Our standard QC measures size and PDI by DLS, and encapsulation efficiency by RiboGreen. Do you have specific requirements for your LNPs regarding size, PDI, and/or EE%? What additional QC you'd like to include?

Upon request there is also the option to measure the zèta potential (electrokinetic potential)
